I believe top players aren't allowed to enter challengers directly so no they're not allowed, however they may get a WC if te tournament were to award it and no challenger tournament in their right mode would turn down a top 5 player for a WC so yeah really they can assuming they get the WC

You are wrong here, TOP10 players aren't allowed to play challengers, not even with a WC.

Thanks for the correction amirbachar. Yes, I finally see it.  To be clear, it is page 44 of an Adobe reader page. It's actually page 86 of the rulebook itself. However, it looks like a player ranked 11-50 may compete in a $50,000 and up Challengers according to the 2nd restriction only by wildcard.
7.07 Play-Up Regulation (ATP Challenger Tour Tournaments)
A. Restrictions1) Players positioned 1-10 in the South African Airways ATP Rankings (singles)
twenty-one (21) days prior to the first Monday of the ATP Challenger Tour tournament
are prohibited from entering, accepting a wild card and/or competing in a
ATP Challenger Tour tournament. Players who would have been, had they and
all other players entered, a direct acceptance on the original acceptance list for a
Grand Slam are prohibited from entering, accepting a wild card and/or competing
in a ATP Challenger Tour tournament in the first week of the Grand Slam tournament.
2) Players positioned 11-50 in the South African Airways ATP Rankings (singles)
twenty-one (21) days prior to the first Monday of the ATP Challenger Tour tournament
are prohibited from entering but may receive an ATP-approved wild card
in order to compete in the Challenger tournament. The Challenger supervisor
will make wild card determinations consistent with the limitations outlined below.
Players positioned 11-50 are also prohibited from entering, accepting a wild card
or competing in ATP Challenger Tour tournaments that offer less than $50,000 in
on-site prize money.

it's sad the Race is already over  Obviously nothing incredible will happen at Shangai and Paris will come too late for anybody to outrank Tipsarevic. He will be #9 whatever happens and alt for Nadal. ATP is still trying to sell it like it is very close and everybody has a chance, but basically it's not
only purpose of the Race is to know who will be #10 by year-end and there are 6 guys fighting for it: Almagro, Monaco, Gasquet, Isner, Raonic and Cilic. And top 12 for AO.

I see those guys are not giving Delpo any penalty for not fulfilling the commitment. Isner, Nadal and Fish also without such penalty. Roddick is dropping way too many points according to them.
Am I missing something ?
Coming week is first with fully automated Masters updates.
Also, if I'm right and live-tennis.eu is wrong about the players I mentioned above, any talk about their site in this thread will be forbidden. If they're right, well, I will make the system better.

The French guy who runs this site and whose pseudo was Chewbacca on MTF made an error, yes : Fed, Nadal, Del Po, Isner, Fish, Roddick, Monfils and Chela will all get a zeropointer, yes, because they have played only 2 ATP500 tournaments so far in the calendar year then for certain they will not be able to fulfill the commitment.
