Björn Borg tried a comeback (I think it was in the early 90:s), with a wooden racquet (!). I think he won 1 match or not even that in two tournaments before quitting tennis once and for all. But can someone come up with a player who has been away from ATP at least say 2 years, but come back and won a title (I know Seles did this in the WTA)?

Mikael Pernfors between the Aus Open 90 and Miami 93 which began at March 8 he played a total of 25 matches due to injuries and won the Canadian Open during 1993.

Muster wasn't away from the tour for 2 years, but that is the best comeback from a serious injury.

What about Sampras before he finally won his last Slam? Or maybe Agassi after his slump?

Oh yes it was. Happy that it turned out to be one of my alltime faves (Pernfors and Kent Carlsson tie as #1).
